چکیده

Magnetic nanoparticles (MNPs) are important contrast agents used to monitor a range of neuropathological processes; microglial cells significantly contribute to MNP uptake in sites of pathology. Microglial activation occurs following most CNS pathologies but it is not known if such activation alters MNP uptake, intracellular processing and toxicity. We assessed these parameters in microglial cultures with and without experimental 'activation'. Microglia showed rapid and extensive MNP uptake under basal conditions with no changes found following activation; significant microglial toxicity was observed at higher particle concentrations. Based on our findings, we suggest that avid MNP uptake by endogenous CNS microglia could significantly limit uptake by other cellular subtypes in mixed neural cell populations.

Iron oxide nanoparticles for neuronal cell applications: uptake study and magnetic manipulations

BACKGROUND The ability to direct and manipulate neuronal cells has important potential in therapeutics and neural network studies. An emerging approach for remotely guiding cells is by incorporating magnetic nanoparticles (MNPs) into cells and transferring the cells into magnetic sensitive units. Recent developments offer exciting possibilities of magnetic manipulations of MNPs-loaded cells by ...

P171: Microglia Cell, Major Player in the Central Nervous System Inflammation

Inflammation, a self-defensive reaction against various pathogenic stimuli, may become harmful self-damaging process. Increasing evidence has linked chronic inflammation to a number of neurodegenerative disorders including alzheimer's disease (AD), parkinson's disease (PD), and multiple sclerosis (MS). In the central nervous system, microglia, the resident innate immune cells play major role in...

Elucidating the Function of Penetratin and a Static Magnetic Field in Cellular Uptake of Magnetic Nanoparticles

Nanotechnology plays an increasingly important role in the biomedical arena. In particular, magnetic nanoparticles (mNPs) have become important tools in molecular diagnostics, in vivo imaging and improved treatment of disease, with the ultimate aim of producing a more theranostic approach. Due to their small sizes, the nanoparticles can cross most of the biological barriers such as the blood ve...

P 155: The Roles of Microglia in Neurodegenerative Diseases

Microglia is a type of glial cell located throughout the central nervous system (CNS), which is sensitive to CNS injury and disease. Responsibility of microglia as the resident macrophage cells for injuries suggests that these cells have the potential to act as diagnostic markers of disease beginning or progression. Function of Microglia is strongly synchronized by the microenvironment of brain...

Poly-l-lysine-coated magnetic nanoparticles as intracellular actuators for neural guidance

PURPOSE It has been proposed in the literature that Fe(3)O(4) magnetic nanoparticles (MNPs) could be exploited to enhance or accelerate nerve regeneration and to provide guidance for regenerating axons. MNPs could create mechanical tension that stimulates the growth and elongation of axons. Particles suitable for this purpose should possess (1) high saturation magnetization, (2) a negligible cy...
